SSR Marker Development and Genetic Identification of Pitaya (<i>Hylocereus</i> spp.) Collected in Okinawa Prefecture, Japan

Pitaya (Hylocereus spp.) is a tropical fruit grown in Okinawa prefecture, Japan. In this study, we developed simple sequence repeat (SSR) markers to assess the genetic diversity of pitaya accessions collected prefecture. SSR were with next-generation sequencing, using Roche 454 GS FLX+ platform. Seventy primer sets tested for identification pitaya, including 50 AG motif and 20 AC SSRs. total, 16 successfully amplified fragments exhibited polymorphism among accessions. The revealed parameters number alleles per locus (ranging from 6 12), expected heterozygosity 0.5 0.85), observed 0.23 0.93), polymorphic information content 0.47 0.83), fixation index value (0.013), out-crossing rate (0.973).
